Abstract :

In the first half of the seminar, it will be presented my previous work on the regulation of m6A RNA modification. In eukaryotes, the interplay between m6A-containing RNA and YTH proteins acts as a fundamental regulatory unit that controls RNA fate, to affect growth and development. However, it is relatively little explored how the m6A-YTH system exerts its regulatory functions in stress adaptation, and much less is known how post-translational modification of YTH proteins can affects on its m6A binding ability. Here, this previous work show that the Arabidopsis YTH protein EVOLUTIONARILY CONSERVED C-TERMINUS PROTEIN 2 (ECT2) is dynamically phosphorylated during heat stress, and further, the ECT2 binding on m6A-containing RNA is affected under high temperature. In the second half, my current project on the plant cell wall integrity mechanisms and oomycete susceptibility will be discussed. In the higher plant Arabidopsis, cell wall integrity (CWI) mechanisms that control tip-growth of root hairs are governed by the MALECTIN-like receptor kinase (MLR) subfamily members, namely FERONIA (FER), and a receptor-like cytoplasmic kinase, namely MARIS (MRI). These pathways are conserved in the tip-growing rhizoids of the basal land plant Marchantia polymorpha and recent omics analyses of Mpfer and Mpmri mutants show a remarkable overlap of the up-regulated genes between these CWI mutants and the wild-type Marchantia infected with the oomycete Phytophtora palmivora. Here, our preliminary results on the responses of Mpfer and Mpmri to P. palmivora infection will be presented.
